| 抄録 | Anticancer drugs induce cell-cycle arrest and apoptosis not only in tumor cells, but also in immune cells. However, many preclinical and clinical findings show that some chemotherapeutic agents can improve the antitumor efficacy of immunotherapy. We immunohistochemically analyzed the degree of immune cell infiltration and the relevance of programmed cell death 1 ligand-1 (PD-L1) expression in surgically resected oral squamous cell carcinoma (OSCC) specimens from patients who had undergone pretreatment with certain chemotherapies and other patients without pretreatment. We divided the patients into the group of neoadjuvant chemotherapy (NAC) patients (n=8) and the nNAC (without NAC) patient group (n=10). We observed that NAC induced infiltrations of CD4, CD8 T cells and CD56 NK cells into the tumor microenvironment. Decreased numbers of Tregs and PD-1-positive cells were observed in the NAC group. No significant difference was observed in the degree of immune-cell infiltration between the patient groups except for CD56 NK cells in the stroma and PD-1 cells in cancer nests. Eighty percent of the nNAC specimens showed intermediate-to-strong PD-L1 protein expression, whereas 75% of the NAC specimens showed down-regulation of the PD-L1 protein, indicating the effectiveness of the chemotherapeutic treatment before surgery. |

| 抄録 | Adenohypophysis delivered from oral ectoderm consists of pars distalis (PD), pars intermedia and pars tuberalis (PT). The mechanisms of development of PD has been well studied, and the cell differentiation of PD has been well understood. However, the morphogenesis and the differentiation of PT are still unclear, and the gene expression during the PT development remains largely unknown. In this study, we explored the specifically expressing genes in PT during development and analyzed its spatiotemporal expressions pattern. Microarray analysis on laser-captured PT and PD tissues obtained from chicken embryos on embryonic day 10 (E10.0) showed high expressing genes, Cytokine-like 1 (CYTL1) and Gap junction protein alpha 5 (GJA5) in PT. A detail analysis of spatiotemporal expressions pattern during chick embryo development by in situ hybridization revealed that CYTL1 mRNA was first detected in lateral head ectoderm and ventral head ectoderm in E1.5. The CYTL1 expressions moved into Rathke’s pouch at E2.5, then it was localized in PT primordium and continuously expressed in PT primordium until E12.0. On the other hand, GJA5 mRNA was transiently detected in PT primordium from E6 to E14.0, while the expression was not detected in PD during development. These results suggested that these genes may be involved in the regulation mechanism of PT development and could be a useful marker in the PT development. |

| 抄録 | In spite of the importance of endocochlear DC potential (EP) and the K+-rich endolymph for the avian cochlea, the structure and function of the tegmentum vasculosum (TV) has not yet been fully elucidated, compared with those of the stria vascularis in the mammalian cochlea. In this study, we examined structural analysis of the epithelial cells, gene expressions of Na+-K+-ATPase (Atp1A1) and Kir4.1 (Kcnj10), and protein localizations of Na+-K+-ATPase and Kir4.1, in the TV. Tight junctional structures were observed between epithelial dark cells and light cells in the TV. Both Atp1a1 and Kcnj10 genes were detected in the TV. In addition, immunopositive signals for both Na+-K+-ATPase and Kir4.1 were recognized in the TV. These results indicate that Na+-K+-ATPase and Kir4.1 play roles in maintaining the EP and high K+ concentration of the endolymph. Further studies are needed to clarify the physiological functions of the TV. This is the first report which demonstrates that gene and protein expression data contribute to the avian inner ear homeostasis. |

| 抄録 | Determination of stem cell hierarchy/lineage is indispensable for a better understanding and augmentation of many aspects of medical sciences, including the mechanisms of tissue development and maintenance of tissue homeostasis, as well as disease development. It also has implications in the field of tissue regeneration for medical treatments and disease modeling for drug discovery using iPS technology. Mesenchymal stem cells are multipotent stem cell that can differentiate into various type of cells including osteoblasts, adipocytes, myocytes and chondrocytes. Runt-related transcription factor 2 (Runx2) is an essential transcriptional regulator of osteoblast differentiation. Runx2 deficiency in Prx1+-derived cells (Runx2prx1−/− mice) resulted in defective intramembranous ossification. Double-positive cells for Prx1-GFP and stem cell antigen-1 (Sca1) (Prx1+Sca1+ cells) in the calvaria expressed Runx2 at lower levels and were more homogeneous and primitive as compared with Prx1+Sca1− cells. Our results suggest that osteoblast differentiation in vivo may begin at the Prx1+Sca1+ MSC stage with sequential progression to Prx1+Sca1−cells, then Osx+Prx1−Sca1− osteoblast precursors, which eventually form mature α1(I)-collagen+ osteoblasts. |

| 抄録 | We report the case of a 58-year-old Japanese man with bilateral synchronous renal cell carcinoma (RCC). The diameters of the right and left tumors were 56 and 69 mm, respectively. Both tumors were endophytic. Cryoablation with prophylactic embolization was performed for the left tumor, and 1 month later, a right open partial nephrectomy was performed. No recurrence was observed during a 16-month follow-up, and the serum creatinine level has been stable. The prognosis of bilateral synchronous RCC is better than that of dialysis patients. The novel approach of combining cryoablation and partial nephrectomy can achieve good cancer control and renal function in bilateral RCC. |

| 抄録 | We experienced a case of advanced maternal age in which a fetus was found to be positive for trisomy 18 at re-examination following indeterminate non-invasive prenatal genetic testing (NIPT), the amniotic fluid chromosomal test revealed a normal karyotype, and confined placental mosaicism (CPM) was observed in an SNP microarray analysis of the placenta. The child was born with no defects or complications. In the present case, the result of the original NIPT at week 15 of pregnancy was indeterminate and the subsequent re-examination result was positive; since the definitive normal diagnosis was not reported until the latter half of week 21, the pregnant patient was subjected to psychological stress for a long period of time. The problem with NIPT is that most of the fetus-derived cell-free DNA in the maternal blood is not derived directly from the fetus but from the villus cells of the placenta, leading to indefinite diagnoses; for that reason, the pregnant patient was subjected to psychological stress for a long period of time. Of the 18,251 cases undergoing NIPT in the past 2 years in Japan, 51 had indeterminate results; this was the second case in which a subsequent re-examination gave a positive result for trisomy 18. |
