抄録 Some mathematical models describing interaction of virus and cells in vivo are reviewed. Similar models using systems of ordinary differential equations can be used for the analysis of dynamics of virus and cells for different kinds of virus. Models for human immunodeficiency virus, hepatitis C virus and hepatitis B virus are treated here. Although models are similar, different approximations can reduce the systems to the explicitly solvable forms. The solutions obtained here can be used to estimate biological parameters.

抄録 Simple mathematical models are considered to explain the pathogenesis of viral hepatitis. Dynamics of populations of liver cells and two virus strains are analyzed qualitatively. This analysis suggests the possibility that the viral mutation causes the hepatitis from the state of carrier.

抄録 Dot-immunobinding assay (DIA) on nitrocellulose membranes and rapid immunofilter paper assay (RIPA) were examind for their usefulness in the detection of cymbidium mosaic virus (CyMV) in orchids. The minimum detection levels of CyMV by these methods were 100 ng/ml in purified preparations and at 10-4 dilution of extracts from infected leaves of orchids could be detected by these methods. Although DIA took 5 to 6 hours for the detection of the virus, it was reliable method for diagnosis of a large-number of samples. On the other hand, RIPA, which enabled detection of CyMV within a few minutes with sensitivity similar to that of DIA, will be suitable as a rapid and handy tool for virus disease diagnosis in orchids. Moreover, by RIPA, we could detect CyMV and odontoglossum ringspot virus (ORSV) simultaneously form doubly infected plant.

抄録 Symptoms on Cymbidium, double-stranded (ds) RNA pattern and peptide mapping of coat protein (CP) of five isolates of odontoglossum ringspot virus from Cymbidium in Japan were compared. One of the isolates, Cy-1, that produced unique symptoms on Cymbidium, showed a distinct peptide mapping pattern from those of the other four isolates by partial digestion of CP with pepsin. All the isolates produced three major dsRNA species of Mr=4.3,1.4 and 0.6×106 in the infected plants.

抄録 To detect cucumber mosaic virus (CMV), virus samples and conjugate were incubated together in a simplified double-antibody sandwich ELISA. The use of the same monoclonal antibody (MAb) as trapping (coating) and detecting antibodies resulted in considerable decrease of ELISA values and sensitivity due to the competition for antigen between trapping and detecting antibodies. The simplified ELISA using two MAbs which recognize different epitopes of CMV proved to be a rapid and sensitive method for virus detection.

タイトル(別表記) ジゴキシゲニン標識プローブを用いたBeet Necrotic Yellow Vein Virus RNA の検出

抄録 Complementary DNA (cDNA) clones corresponding to each of five distinct RNA species of beet necrotic yellow vein (BNYVV) were synthesized and identified. The sizes of each cDNA clone for RNAs 1,2,3,4 and 5 molecules were 3.0, 1.7, 1.8, 1.5 and 1.4 kbp, respectively. cDNA inserts to RNA 2 were covered at a part of the 3'regions, and those of RNAs 3,4 and 5 were almost full-length. The plasmids containing each of cDNA inserts were labeled with digoxigenin by the random priming method. Northern blot hybridization tests showed that individual probes hybridized specially to each of the five RNAs. Good results were obtained with 1 to 10 ng of RNA as a mixture of five RANs, but the probe to RNA 3, RNA 4 or RNA 5 gave a weak signal with hererologous RNAs when more than 10 ng RNA was used. In dot blot hybridization, the limit of detection was about 10 pg RNA, but if a higher content of RNA was spotted, cross reaction occurred using heterologous RNAs. For laboratory and field isolates of BNYVV, each of RNAs 3,4 and 5 was easily detected by Northern blot hybridization in total nucleic acids extracted from Tetragonia expansa leaves inoculated mechanically, but not from roots of sugar-beet plants inoculated by the fungus Polymyxa betae. However, satisfactory results were obtained with partially purified or concentrated preparations from roots. These findings indicate that the digoxigenin-labeled probes are useful for the identification and detection of RNAs contained in field and laboratory isolates of BNYVV.

抄録 Bean yellow mosaic virus (BYMV) was isolated from Calanthe sp.showing mild chlorosis on the leaves, collected in Yamaguchi Prefecture in Japan. The virus was transmitted by the aphid Myzus persicae in a non-persistent manner, and by sap-inoculation to 29 out of 46 plant species from 9 out of 12 families tested. Stability in crude sap, morphology of virus particles, shape of cylidrical inclusions and the presence of cytoplasmic crystalline inclusions in the infected cells were similar to those of BYMV isolates previously reported. The virus contained a single protein species with a molecular weight of 35,000. In a microprecipition test and double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), the virus showed a close serological relationship to isolates of BYMV from both crocus and gladiolus, and showed a distant relationship to clover yellow vein virus. Three BYMV isolates used in this study were found to be serologically related to each other, but the virus was more closely related to the BYMV crocus isolate than to gladiolus isolate.

抄録 A potyvirus, isolated from Habenaria radiaata showing severe mosaic in the leaves named habenaria mosaic virus(HaMV). HaMV was transmitted by inoculation of sap to 6 of 41 species from 4 of 12 families. Systemic infection was cauded in Habenaria radiata, and local infection was produced in Chenopodium amaranticolor, C. quinoa, Spinacia olracea, Tetragonia expansa and Gomphrena globosa. HaMV was also transmitted by Myzus persicae in a non persistent manner, but not by Aphis gossypii. The virus has filamentous particles of c. 750×13nm. Sap from inoculated C. quinoa was infective after dilution to 10-5 but not 10-6, after 10 min at 60℃ but 65℃, or after 8-16 days at 20℃.In ultrathin sections of diseased leaves of Habenaria radiata and inoculated C. quinoa, virus particles were found to disperse in the cytoplasm. Cylindrical inclusions were present in the cytoplasm of infected cells, as pinwheels, scrolls or tubes. The inclusions observed in negatively stained crude sap preparations exhibited fine linear striation at about 5.2 nm intervals.The antiserum to HaMV produced by immunizing a rabbit had a titer of 1/512(precipitin test). HaMV showed distant serological relationships to potyvirus;alstroemeria mosaic virus, bean yellow mosaic virus, calanthe mild mosaic virus, clover yellow vein virus, dendrobium mosaic virus, freesia mosaic virus, turnip mosaic virus and watermelon mosaic virus 2.

抄録 Azuki bean mosaic virus (ABMV) was isolated from Glycine max showing mild mosaic and stunt on the leaves, collected in Ibaraki-Prefecture, in July, 1989. ABMV was transmitted by sap-inoculation to 8 species systemically and 14 species locally out of 33 species in 8 families and by aphids, Aulacorthum solani and Myzus persicae in a non-persistent manner.Seed transmission was recognized in 0.8% of the seeds of soybean cv."Iwate-wase-kurome" inoculated at seeding stage. The soybean plant infected with ABMV produced mottled seeds, which were radical- or saddle-like. The virus particles were flexuous rods, bout 750 nm in length. In ultrathin virus-infected tissues, cytoplasmic inclusions containing pinwheels and scrolls were observed in the cytoplasms. In DAS-ELISA, the virus reacted strongly with antiserum to ABMV. The weight of plants and seed of two soybean cv."Okuhara 1" and "Iwate-wase-kurome" inoculated with ABMV at the seedling stage was about 40~50% less than that of healthy plants.

抄録 A strain (Ixia-B) of bean yellow mosaic virus (BYMV) isolated from Ixia hybrida was characterized and compared with other isolates of BYMV and clover yellow vein virus (CYVV). Ixia-B was transmitted by aphids,Myzus presicae in a non-presistent manner and by sap-inoculation to 11 of 46 species in 5 of 10 families tested, and had a similar host range to that of some BYMV isolates, althrough some defferences were detected. Sap from diseased C. quinoa was infective after 10 min heating at 55℃ but not 60℃, after a dilution to 10-3 but not 10-4, and after 2 days but not 4 days at 20℃.The Virus particles were filamentous rods of about 13×820 nm. Ixia-B contaied a single protein species with a molecular weight of 34,000 and a single viral RNA with approximately 9,000 bases. In ultrahtin sections of leaf tissues from infected plants, the virus particles, cylindrical cytoplasmic inclusions and dense bodies were obsserved in the cytoplasm. The antiserum to Ixia-B produced by immunizing a rabbit had a titer of 1/512. A close serological relationship was revealed between Ixia-B and two strains of BYMV from crocus and gladiolus, but no relationship to clover yellow vein virus was found in agar gel diffusion tests. However,Ixia-B could be distinguished from two strains of BYMV by the formation of spurs among them in agar gel and by the differences in the patterns of peptide mapping of coat proteins. From these findings, Ixia-B was identified as a strain of BYMV.
