kn-abstract=本研究の目的は,性的マイノリティ当事者の語りが,聴講者の性的マイノリティへの態度や認識に及ぼす変化を明らかにすることである.当事者が現職の学校教員と校長を対象に50分の講話をし,講演の前後に質問紙調査を行い,印象や当惑感,理解の困難さについて数値化し,変化を調べた.調査内容は,印象の測定にはSD法を使用し,印象の「身近さ」因子に含まれる6項目について7件法で回答を求め,当惑感については8項目,理解困難については2項目の尺度を使用し,6件法で回答を求めた.その結果,参加者は性的マイノリティをより身近に感じるようになり,当惑感や理解困難等の否定的な態度や認識が有意に軽減した. In the present study, we focused on the SIgA coating of gut bacteria as a mucosal immune response affecting the gut bacteria following a high-fat diet (HFD). This study newly showed that the level of SIgA coating of gut bacteria was significantly decreased in HFD-fed mice than normal-fat diet (NFD)-fed mice. Furthermore, we found that the relative abundances of gut bacteria were significantly negatively or positively correlated with the level of SIgA coating of gut bacteria in NFD-fed and HFD-fed mice. Melanocyte precursors have high mobility and migrate and settle into various tissues and organs, including the skin, choroid, inner ear, heart, brain, adipose tissue, lung, etc. It is well known that RPE cells are essential for visual acuity and that melanocytes in the skin provide protection against UV-damage. So, what is the function of extracutaneous melanocytes that are distributed elsewhere in the body where only very small amounts of light illuminate their existence? Here I would like to briefly introduce our research history and then discuss the important roles of melanocytes in the inner ear that are indispensable for hearing ability and those in the choroid Eimeria parasite is causative agent of this disease. In Japan, this parasite species were widely spread and the positive rates are about 50 in layer and 70 % in broiler. The symptoms of coccidiosis are diarrhea, bloody excretion, weight loss, and die. Eimeria tenella is the most pathogenic protozoa. The sporozoites, infection form of this parasite, entry to epithelial cells around the crypt of cecum in early infection. After infection, parasites proliferate in epithelial cells, and form to sexual stage finally. However, we have less information about the pathophysiology, especially invasive mechanisms and infection route, by E. tenella infection. We have focused to analyze the invasive mechanism and route of this parasite because this phenomenon is first event to cause the pathophysiological changes in the infection. Mammalian hibernators store fat extensively in white adipose tissues (WATs) during pre-hibernation period to prepare for hibernation. Thus, they undergo adaptive body remodelling in autumn, the pre-hibernation period, prior to hibernation, whereas little is known about the physiological and molecular mechanisms of the pre-hibernation remodelling. Syrian hamsters (Mesocricetus auratus) are facultative hibernators that can hibernate irrespective of seasons when exposed to prolonged short photoperiod and cold ambient temperature (SD-Cold) conditions. We found that before the initiation of hibernation, the body mass of Syrian hamsters decreased below a threshold, indicating that hibernation in this species depends on body condition. Global profiling of gene expression in various tissues of animals during the pre-hibernation period and hibernation period identified adaptive changes for hibernation at molecular level. After injection of metallic mercury into mice, the mice that received the incomplete cubane-type sulfur-bridged nitrilotriacetato molybdenum (IV) complex ([Mo3S4(Hnta)3]2-, referred to as the NTA complex) had the highest mercury content in their urine on the first day, followed by L-cysteine solution and then water. On the other hand, after injection of mercuric ion, the urinary mercury excretion from the mice that received various concentrations of the incomplete cubane-type sulfur-bridged cysteinato molybdenum (V) complex ([Mo2O2S2(cys)2]2-, referred to as the CYS complex) showed a higher level than that of mice that received water (control group). The mercury content in the heart and lungs of the group that received the CYS complex solution decreased in comparison with that of the group that received the water. Ovarian transplantation experiment suggested that lack of ovulation in Runx3−⁄− mice was caused by alteration of gonadotropin secretion in Runx3−⁄− mice. Cyp11a1 mRNA expression was less in Runx3−⁄− mouse ovaries than in wt ones. Hypothalamic Gnrh1 mRNA was increased, and Kiss1 mRNA expression in the anteroventral periventricular nucleus was decreased, but Kisspeptin mRNA in the arcuate nucleus was increased in Runx3-/- mice. Pituitary Fshb mRNA levels were increased in Runx3−⁄− mice. Cholesterol side-chain cleavage enzyme gene (Cyp11a1) expression was decreased in ovaries of Runx3−⁄− mice. These findings suggest that anovulation in Runx3−⁄− mice was partly due to the alterations in hypothalamus-pituitary-ovary system. Parents of pre-school children, aged 3-5 years and/or 1st and 2nd graders answered a self-reported questionnaire on child rearing. This data of 166 parents were analyzed qualitatively. Most parents with children before and after entering elementary schools reported happiness through child growth and being together. On the other hand, some parents felt anxiety concerning child adjustment in school along with their own attitudes toward their children. It was suggested that collaborative child care support during pre-school and elementary school years would be helpful in providing opportunities to facilitate communication between other parents. This would include consultation concerning child rearing anxiety, and opportunities to attend lectures and psychoeducational workshops. Evaluation of child care support systems is necessary for better outcomes. It also has implications in the field of tissue regeneration for medical treatments and disease modeling for drug discovery using iPS technology. Mesenchymal stem cells are multipotent stem cell that can differentiate into various type of cells including osteoblasts, adipocytes, myocytes and chondrocytes. Runt-related transcription factor 2 (Runx2) is an essential transcriptional regulator of osteoblast differentiation. Runx2 deficiency in Prx1+-derived cells (Runx2prx1−/− mice) resulted in defective intramembranous ossification. Double-positive cells for Prx1-GFP and stem cell antigen-1 (Sca1) (Prx1+Sca1+ cells) in the calvaria expressed Runx2 at lower levels and were more homogeneous and primitive as compared with Prx1+Sca1− cells. As a result, broiler chickens grow faster than layer chickens. However, excessive accumulation of body fat in broiler chickens has been a serious problem in the poultry industry in recent decades. Therefore, the appetite regulatory system of chickens has been a focus of research among poultry scientists. Lines of evidence suggest that the physiological role of peripheral adiposity hormones, such as leptin and insulin, and gut hormones, such as ghrelin, are different between mammals and chickens. Thus, species specificity make it difficult to understand the mechanisms underlying the regulation of food intake in chickens. The detailed MC of the rat was examined, using inulin as a marker of the applied solution. Inulin disappeared monoexponentially from the nasal cavity, indicating that the MC can be assumed to follow first-order kinetics. In the proposed kinetic model, the fractional absorption of the drug following nasal application is predicted as the sum of fractional absorption from the nasal cavity and fractional absorption from the GI tract, both of which are estimated indirectly from the permeability to the Caco-2 monolayer. This kinetic model is the first estimation system for nasal drug absorption based on drug disposition after nasal application and is useful for the development of nasal dosage forms. en-copyright= To avoid that the inconsistency becomes any problems, Okayama University has been established new centralized animal facility to propel the institutional management. Secreted Frizzled-related protein 1 (Sfrp1) is highly expressed in kidney, although little is known about connection between the protein and renal diseases. Here we focused on Sfrp1 to investigate its roles in renal fibrosis using a mouse model of unilateral ureteral obstruction (UUO). In wild-type mice, the expression of Sfrp1 protein was markedly increased after UUO. The kidneys from Sfrp1 knockout mice showed significant increase in expression of myofibrobast markers, alpha-smooth muscle actin (SMA). Sfrp1 deficiency also increased protein levels of the fibroblast genes, Vimentin, and decreased those of the epithelial genes, E-cadherin, indicated that enhanced epithelial-to-mesenchymal transition (EMT). There was no difference in the levels of canonical Wnt signaling; rather the levels of phosphorylated c-Jun and JNK were more increased in the Sfrp1-/- obstructed kidney. Moreover, the apoptotic cell population was significantly elevated in the obstructed kidneys from Sfrp1-/- mice following UUO, but was slightly increased in those from wild-type mice. Sex determination mechanism is varied depending on the species, however, sex differentiation mechanism in vertebrates are basically similar. Sex determination of birds is controlled by the sex chromosomes and their sexual differentiation are critically controlled by estrogen. Because estrogen is biosynthesized by aromatase, it is possible to find the sex-determining gene of birds by analyzing the transcriptional regulatory mechanism. While there have been conducting research on the basis of this concept, the mechanism has not yet been fully elucidated. On the other hand, DMRT1 gene has been cloned as a strong candidate in the sex-determining gene of birds. The information is transmitted to the cochlear nucleus in the brainstem via spiral ganglion neurons. The information is further transmitted to higher relaying centers in the brain such as superior olivary complex and inferior colliculus. The connectivity between these components is topographically organized in a frequency-specific manner. It is known that the organization is well-established from the beginning of the circuit development. However, little is still known about the molecular mechanisms underlying the development of connectivity in the auditory circuit. Homeobox transcription factors of the Hox gene family are known for their involvement in early anterior-posterior axis patterning of neuronal progenitors in the hindbrain. Recent evidence indicates that they also play important roles in late aspects of neuronal development and establishment of topographic circuitry. Moreover, a mutation in the HOXA2 gene has been recently shown to be responsible for hearing deficits in humans. Orally administered acetate is readily absorbed in the blood stream and then taken up by tissues and activates AMP-activated protein kinase (AMPK) by increasing the AMP/ATP ratio. Administered acetate shows a marked reduction in lipid accumulation in the adipose tissue, protection against accumulation of fat in the liver, and improves glucose tolerance. It decreases the transcripts of the lipogenic genes in the liver, indicating an inhibition of lipogenesis in that organ. Furthermore, acetate treatment shows a higher rate of oxygen consumption and a smaller size of lipid droplets in white and brown adipose tissues. Recently, the mutation of WW domain containing E3 ubiquitin protein ligase 1 (WWP1) gene has been identified as a responsible for muscular dystrophy chicken. We observed that muscular dystrophy chicken not only showed the degeneration of skeletal muscles but also produced watery feces. Therefore, we examined the possibility of abnormalities in water metabolism of muscular dystrophy chicken. We first analyzed plasma osmolality and gene expression of aquaporin 2 (AQP2), AQP3 and alpha subunit of the amiloride-sensitive epithelial sodium channel (αENaC) in muscular dystrophy chicken and White Leghorn chicken under normal physiological conditions at five-week old. Subsequently, we analyzed these same parameters after one-day water-deprivation. The main findings of our study are that: I) the plasma osmolality was significantly higher in muscular dystrophic chicken than in White Leghorn; II) kidney αENaC mRNA expression was significantly lower in muscular dystrophic chicken than in White Leghorn; III) AQP2 and AQP3 mRNA expressions in muscular dystrophic chicken were similar in White Leghorn. This transition, known as an oocyte-to-embryo transition, is critical step for further embryonic development. Considering that the process occurs quickly, bulk degradation system could be highly activated to eliminate maternal products and recycle them to synthesize newly products. We have demonstrated that autophagy, which is cytoplasmic bulk degradation system mediated by the lysosome, is highly induced after fertilization, and autophagy-deficient embryo dies before implantation, suggesting that autophagy is essential for preimplantaion embryonic development. Our recent studies also revealed that lysosomal size and number changes during the embryonic development. problem. Many patients with DM in Asia, South America, India and East Africa have traditionally used the water extract of unripe fruits of Momordica charantia (bitter melon) as some form of complementary and alternative medicine. Studies of laboratory animals have shown the beneficial blood-glucose lowering and anti-diabetic effects of this remedy. Some oral components that bring lower blood glucose level have been isolated : charantin (sterol glycosides), charantin (polypeptide) and cucurbine-type triterpenes. Part of their actions are related to AMP-activated kinase and repression of the oxidative stress that is increased in DM. Most clinical reports are not fully convincing due to the lack of randomized control studies. The present article reviews the pharmacological and clinical effects of bitter melon with special emphasis on the anti-diabetic effects, and some effects that would require caution in the context of In Edo era (1603-1868), male quails were housed in Samurai class as a pet of "Singing bird" to encourage his mind. After that, these birds were domesticated for poultry under the policy of Meiji government, JAPAN. However, the Japanese quail as an experimental animal was lately paid attention in the U.S.A after 1950. Since 1960 in Japan, the Japanese quail has been improved for an experimental animal by using the domesticated ones. The National Institute for Environmental Studies (NIES) has exploited the Japanese quail as a useful experimental animal for environmental science by cooperating with Tohoku University since 1980. By the way, the Japanese quail (Coturnix japonica) was selected for high (H) and low (L) antibody production to Newcastle disease virus (NDV) vaccine more than 50 generations. And H and L quails had been reached the plateau of NDV-HI (Haemo-agglutinin Inhibition) titer at 24(th) generation. The structure of major histocompatibility complex (MHC) genes of the H and L quails was examined and found that each line was independent MHC homozygous line. During the selection experiment, these H and L quails were examined in their utilization as an experimental animal for Environmental Science as follows: 1) The reproductive traits such as egg weight, egg production, hatchability declined in H and L line quails, and H line became extinct at 53 generation due to inbreeding depression. However, L line has been bred as inbred quail for avian experimental animal, because of hatchability recovery happened at 43 generation. 2) Among the 4 species (mice, rat, hamster, quail), the Japanese quail showed highest sensitivity to NO(2). On the contrary, the sensitivity to O(3) was lowest in quails. It has been suggested that toxic mechanisms of those gases are different from one another. 3) The environmental hormone (Endocrine disrupting chemicals, EDCs) was examined in the hybrid egg of quails in accordance with the OECD guideline. These results demonstrate that EDCs such as DDT, Dieldrin and TBTO will damage the reproductive organs and make the egg shells thin in adult female. rhythm generation kn-title=コオロギを実験動物とした生物時計の解析: 時計遺伝子periodのリズム発現機構における機能解析 en-subtitle= kn-subtitle= en-abstract= kn-abstract=フタホシコオロギでは、分子生物学的手法が開発されつつある。ここでは、最近われわれが試みているRNA干渉による、フタホシコオロギ(Gyyllus bimaculatus)の時計遺伝子perの機能解析を紹介した。幼虫頭部での解析により、per mRNAの発現量は、夜の始めにピークをもつリズムを示し、このリズムは恒暗・恒温条件下でも継続することが明らかとなり、perの時計機構への関与が示唆された。そこで、per dsRNAを用いたRNA干渉により、per遺伝子の役割を検討した。幼虫へのper dsRNAの投与により、per mRNAレベルは対照群の25%にまで減少し、かつほとんどの個体で活動リズムが消失することがわかった。これらの結果はコオロギでもperがリズムの発現に重要な役割を担うことを示唆している。さらに、終齢幼虫にper dsRNAを投与した場合も、羽化後の活動が恒暗条件下で無周期となることがわかった。これらの結果から、コオロギではperが時計機構に必須であることが示唆された。また同時に、RNA干渉が時計遺伝子の機能解析に極めて有効な手段であることが確認された。 en-copyright= kn-copyright= en-aut-name=SakamotoTomoaki en-aut-sei=Sakamoto
