UniversityThe postharvest properties of two ultra-late maturing peach cultivars, "Tobihaku" (TH) and "Daijumitsuto" (DJ), were investigated. Fruit were harvested at commercial maturity and held at 25 degrees C. TH exhibited the characteristics of normal melting flesh (MF) peach, including rapid fruit softening associated with appropriate level of endogenous ethylene production In contrast, DJ did not soften at all during 3 weeks experimental period even though considerable ethylene production was observed. Fruit of TH and DJ were treated with 5,000 ppm of propylene, an ethylene analog, continuously for 7 days. TH softened rapidly whereas DJ maintained high flesh firmness in spite of an increase in endogenous ethylene production, suggesting that DJ but not TH lacked the ability to be softened in response to endogenous and exogenous ethylene/propylene. DNA-seq analysis showed that tandem endo-polygalacturonase (endoPG) genes located at melting flesh (M) locus, Pp-endoPGM (PGM), and Pp-endoPGF (PGF), were deleted in DJ. The endoPG genes at M locus are known to control flesh texture of peach fruit, and it was suggested that the non-softening property of DJ is due to the lack of endoPG genes. On the other hand, TH possessed an unidentified M haplotype that is involved in determination of MF phenotype. Structural identification of the unknown M haplotype, designated as M-0, through comparison with previously reported M haplotypes revealed distinct differences between PGM on M-0 haplotype (PGM-M-0) and PGM on other haplotypes (PGM-M-1). Peach M haplotypes were classified into four main haplotypes: M-0 with PGM-M-0; M-1 with both PGM-M-1 and PGF; M-2 with PGM-M-1; and M-3 lacking both PGM and PGF. Re-evaluation of M locus in association with MF/non-melting flesh (NMF) phenotypes in more than 400 accessions by using whole genome shotgun sequencing data on database and/or by PCR genotyping demonstrated that M-0 haplotype was the common haplotype in MF accessions, and M-0 and M-1 haplotypes were dominant over M-2 and M-3 haplotypes and co-dominantly determined the MF trait. In this study, we attempted to develop a VIGS-based gene evaluation system for seven Prunus species, including apricot (P. armeniaca L.), sweet cherry (P. avium L.), almond [P. dulcis (Mill.) D. A. Webb.], peach (P. persica Batsch), Japanese apricot (P. mume Siebold & Zucc.), Japanese plum (P. salicina Lindl.), and European plum (P. domestica L.), with the Apple latent spherical virus (ALSV) vectors. ALSV vectors carrying part of the apricot PHYTOENE DESATURASE (PDS) gene sequence were amplified in Nicotiana benthamiana, and inoculated into the cotyledons of Prunus seedlings by particle bombardment. Typical PDS-silenced phenotypes, characterized by uniform discoloration of the upper leaves, were observed in sweet cherry and some cultivars of apricot and almond several weeks after inoculation. In contrast, attempted ALSV infections of Japanese apricot, Japanese plum, European plum, and the other cultivars of apricot and almond were unsuccessful. Furthermore, although the infection rate of ALSV in peach was high, severe viral infection symptoms were observed in the infected leaves. These results collectively suggested that the efficiency of ALSV infection and VIGS could vary depending on species and/or cultivar in Prunus. isolated from Calanthe sp.showing mild chlorosis on the leaves, collected in Yamaguchi Prefecture in Japan. The virus was transmitted by the aphid Myzus persicae in a non-persistent manner, and by sap-inoculation to 29 out of 46 plant species from 9 out of 12 families tested. Stability in crude sap, morphology of virus particles, shape of cylidrical inclusions and the presence of cytoplasmic crystalline inclusions in the infected cells were similar to those of BYMV isolates previously reported. The virus contained a single protein species with a molecular weight of 35,000. In a microprecipition test and double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), the virus showed a close serological relationship to isolates of BYMV from both crocus and gladiolus, and showed a distant relationship to clover yellow vein virus. HaMV was transmitted by inoculation of sap to 6 of 41 species from 4 of 12 families. Systemic infection was cauded in Habenaria radiata, and local infection was produced in Chenopodium amaranticolor, C. quinoa, Spinacia olracea, Tetragonia expansa and Gomphrena globosa. HaMV was also transmitted by Myzus persicae in a non persistent manner, but not by Aphis gossypii. The virus has filamentous particles of c. 750×13nm. Sap from inoculated C. quinoa was infective after dilution to 10-5 but not 10-6, after 10 min at 60℃ but 65℃, or after 8-16 days at 20℃.In ultrathin sections of diseased leaves of Habenaria radiata and inoculated C. quinoa, virus particles were found to disperse in the cytoplasm. Cylindrical inclusions were present in the cytoplasm of infected cells, as pinwheels, scrolls or tubes. The inclusions observed in negatively stained crude sap preparations exhibited fine linear striation at about 5.2 nm intervals.The antiserum to HaMV produced by immunizing a rabbit had a titer of 1/512(precipitin test). ABMV was transmitted by sap-inoculation to 8 species systemically and 14 species locally out of 33 species in 8 families and by aphids, Aulacorthum solani and Myzus persicae in a non-persistent manner.Seed transmission was recognized in 0.8%
of the seeds of soybean cv."Iwate-wase-kurome" inoculated at seeding stage. The soybean plant infected with ABMV produced mottled seeds, which were radical- or saddle-like. The virus particles were flexuous rods, bout 750 nm in length. In ultrathin virus-infected tissues, cytoplasmic inclusions containing pinwheels and scrolls were observed in the cytoplasms. In DAS-ELISA, the virus reacted strongly with antiserum to ABMV. The virus was transmitted by sap inoculation to 12 of 57 species in 6 of 12 families tested, but not by aphid Mizus persicae or through seeds. Systemic infection occurred in all Orchidaceae plants tested and only one in non-orchidaceae (Sesamum indicum). In Tetragonia expansa sap, the infective at a dilution of 10-5 but not at 10-6, after heating at 65℃ for 10 min, and was still active after 1 month aging in vitro. Flexuous rod particles, c. 475×13nm,
were observed.In ultrahtin sections of leaf tissues from diseased plants, virus particles were found to aggregate in the cytoplasm. The molecular weight of the protein submit and RNA determined by gel electrophoresis, was 27.8×103 and 2.2×106, respectively. Double-stranded RNAs with estimated molecular weight of 5.4×106, 4.0×106, 3.6×106 and 3.0×106 were isolated from infected plants.No potential conflict of interest relevant to this article was reported.岡山大学農学部Acta Medica Okayama0474-02548811999モモの生理的落果と果実発育との関連159163ENFumioFukudaIn order to clarify the factors that cause fruit pit-spilitting and physiological fruit drop in peach (Prunus persica cv.'Shimizu-hakuto'),the effects of fruit thinning on fruit development were investigated.In addition,the characteristics of fruit development in 'Shimizu-hakuto'were compared with those in early-ripening cultivars which cause heavy pit-splitting but low physiolofical fruit drop,and in 'Yamao-hakuto'which hardly cause any pit-splitting or physiological fruit drop.Pit-spilit and fallen fruits genarally show a rapid enlargement at the end of stage Tof fruit growth than normal fruits.At completion of pit splitting the percentage dry weight content of the pit reached 30%.
Fruit with relatively less increase in length that of diameter and width,at the early stage of the fuit development,showed low pit-splitting occurrence in all the cultivars.Early in the stage of fruit development, we observed two vascular bundles attached to the seed of the early-ripening cultivars,but only one in 'Shimizu-hakuto'and'Yamato-hakuto'.Embryos in pit-spilit fruit were smaller than in normal fruit,and their length in fallen fruit was less than 5mm.The development of endosperm was also delayed in fallen fruit.No potential conflict of interest relevant to this article was reported.岡山大学農学部Acta Medica Okayama0474-02546111983ユスラウメ台及び共台のモモの生育と収量6775ENMikioNakanoKazuoShimamuraモモ栽培の省力化を目的に台木及び仕立方を異にした低木･密植栽培を試みた. すなわち,ユスラウメ台を用いた主幹仕立,共台を用いた架線仕立と平行並列仕立である. ユスラウメ台樹の生育は品種及び個体により大きく変異したが,共台に比べ強くわい化した. 収穫期は早く,果実の品質も良かった. 収量はほぼ生育と一致し,品種及び個体により大きく変異したが,生育の良い場合には共台樹より多収であった. したがって,台木の系統選抜などにより健全で均一な生育が得られれば,有望な栽培様式となりえよう. 共台樹では平行並列仕立の方が架線仕立より収量,品質とも優れた. その原因は,平行並列仕立の方が目標樹冠面積が大きく,自由に生長させられたのに対し,架線仕立樹はそれが狭く,しかも,主枝を早くから低く誘引したため,樹勢が強すぎ,徒長的な側枝が多発し,過繁茂となったためであろう。No potential conflict of interest relevant to this article was reported.
