Study on the Free and Cojugated Steroids in Normal Placenta

Abstract
従来,内分泌臓器としての胎盤からステロイドホルモンを抽出分離定量している報告は数多くあるが,そのいずれもが単一のステロイドのみを定量したものか,あるいは大量複数の胎盤を使つてのステロイドの抽出分離定量したかのいずれかにすぎなかつた.しかし胎盤組織内でのステロイドの代謝過程が次第に解明されつつある現在単一のステロイドのみ測定や大量複数の胎盤を使つてのステロイド測定の意義は徐々に失なわれつつある.そこで著者は正常満期産1個の胎盤におけるmetabolitesを測定することは胎盤における個体差及びその特徴を知る上に重要と考え,従来の定量法の難点を解決し満期産1胎盤組織中から測定しうる各種ステロイド(21種)を同時に抽出分離精製定量する新定量法を確立した.この方法により正常満期産胎盤16個について遊離型ステロイド21種を測定することができた.更に結合型ステロイドであるGlucuronide, Sulfateについても独自の方法により3胎盤を1つの測定単位として抽出分離定量し遊離型ステロイドと対比した.その結果中性遊離型ステロイドでは活性型以外に5位の飽和された不活性型が相当みとめられ△4-typeと5位の飽和型の比は1:1.7であつた.結合型ステロイドについてはエストロゲンは少量ながらGlucuronide, Sulfate共にみとめられたがP. nolone, 17-OH-P. nolone, DHA+Andr.にはSulfateが認められ,中でもDHA+Andr.では遊離型より多く認められた.Tetrahydro. Comp.のCorticoidにはGlucuronideが認められTHEは遊離型より多量に胎盤中に存在していた.
Abstract Alternative
In this study, the approach was tried with the determination of steroids in placenta. Twenty one steroids in placenta were determined by new method, which was consisted of following procedure. 1) 2 steps hydrolysis (hydrolysis with β-glucuronidase in acetate buffer and solvolysis) 2) Florisil column chromatography 3) Thin layer chromatography The results of this determination were as followings. I. Free steroid in placenta A) Estrogen (μg/one placenta) Estrone: 33.0±:6.9, Estradiol: 37.6±11.7, 16-epi-Estriol: 17.9±2.6, Estriol: 57.6±10.3, Estriol content in placenta was not high as was considered. B) Gestagens (μg/one placenta) P. nolone: 43.8±10.3, 17-OH-P. nolone: 50.2±13.3, Prog.: 36.1±10.6, 17-OH-Prog.: 16.9±5.9 C) Androgens (μg/placenta) ⊿4-A-dione: 23.0±4.0, DHA+Andr.: 8.6±3.8, Etio.: 7.9±2.5, 11-keto-Etio.: 14.6±3.2, 11-OH-Etio.; 14.6±4.6 D) Corticoids (μg/placenta) Less polar subst.: 10.2±1.6, DOC: 6.8±3.5, Comp. A: 3.7±2.3, Comp. B: 1.1±1.1, Comp. E: 17.9±2.8, Comp. F: 13.2±4.2, Comp. S: (一), THS: 3.1±2.8, THE: 13.9±4.6, THF: 35.0±5.8, More polar subst.: 13.0±5.8 II. Cojugated steroids in placenta A) Estrogen (μg/placenta) Estrogen as sulfate were present one tenth of free type in placenta. There were a little content of 16-epi-Estriol and Estriol glucuronide. B) Gestagens (μg/placenta) P. nolone and 17-OH-P. nolone that were ⊿5-3β-ol type steroids were estimated as sulfate. C) Androgens DHA+Andr. fraction as sulfate was estimated higher content than free type. All androgens glucuronide were not meassurable in placentar tissue. D) Corticoids Tetrahydro. comp. that were THF, THE, THS etc, as glucuronide were estimated considerably high. Above all, THE glucuronide was estimated higher content than free type of THE.
